HeLa cells (low-expressing cell line, panel A) and U87-MG cells (positive cell line, panel B) were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-CD73 (clone W23165G), followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405405) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 40X objective and merged. Scale bar: 50 µm -
Human peripheral blood lymphocytes were stained with PE anti-human CD19 (clone HIB19, Cat. No. 302207) and CD73 Purified (clone W23165D) (left) or Purified rat IgG2a, κ (clone RTK2758, Cat. No. 400501) Isotype Control (right), followed by Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416).

CD73 (cluster of differentiation 73) is a membrane-bound glycoprotein and ectoenzyme. To activate adenosine signaling CD73 catalyzes the hydrolysis of AMP generated by CD39 to adenosine. This role makes CD73 crucial to the regulation of purinergic signaling and immune homeostasis and response. CD73 also serves a variety of physiological functions including lymphocyte binding, epithelial ion transport, ischemic preconditioning, tissue-barrier function, T cell activation, and tissue homeostasis. Overexpressed in several cancers and implicated in tumor progression, metastasis, and the impairment of anti-tumor immune response via adenosine signaling, CD73 is a target of interest in cancer therapy. It is also a potential immunotherapy target for the treatment of pain, inflammation, and hypoxia.

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.03 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

This product is not recommended for use in immunohistochemistry in formalin fixed, paraffin-embedded tissues (IHC-P) or Western blot (WB).
