Whole cell extracts (15 µg total protein) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-GLI3 (clone W24017B) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control. Description

GLI3 is a transcription factor in which plays a significant role in embryonic development and cellular differentiation. It belongs to the GLI family of zinc-finger transcription factors, which mediate the Hedgehog (Hh) signaling pathway. One of the key functions of GLI3 is in limb and brain development, where it regulates gene expression patterns crucial for proper morphogenesis. In the absence of Hedgehog signaling, GLI3 is processed into a truncated repressor form (GLI3R), which suppresses the transcription of Hedgehog target genes. In the presence of Hedgehog signaling, GLI3 exists in a full-length activator form (GLI3A), promoting gene expression. This dynamic regulation is essential for processes such as digit specification in limb formation, as GLI3 controls the balance between anterior and posterior limb identities.

Mutations in GLI3 are linked to several congenital disorders, including Greig cephalopolysyndactyly syndrome (GCPS), Pallister-Hall syndrome (PHS), and polydactyly. GLI3 also plays a role in neural tube patterning, contributing to the differentiation of neurons and glial cells which involves other signaling pathways, such as Wnt and BMP, highlighting its integrative role in developmental regulation. Beyond embryogenesis, GLI3 is implicated in cancer progression and tissue homeostasis, as aberrant Hedgehog signaling and GLI3 mutations have been associated with tumorigenesis in basal cell carcinoma and medulloblastoma.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 0.5 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use of this antibody in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) or True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401). Fixation/permeabilization with True-Phos™ Perm Buffer (Cat. No. 425401) is not recommended.

This antibody is not recommended for immunocytochemistry (ICC) or immunohistochemistry in FFPE tissue (IHC-P).
