IHC staining with Purified anti-Keratin14 (clone W24068C) on formalin-fixed paraffin-embedded human skin tissue. Following antigen retrieval using Sodium Citrate H.I.E.R. (Cat. No. 928502), the tissue was incubated with primary antibody overnight at 4°C. The next day, tissues were incubated with Biotin Goat anti-rat IgG Antibody (Cat. No. 405428), followed by BioLegend’s Ultra Streptavidin HRP Kit (Multi-Species, Cat. No. 929501) and AEC Chromogen Concentrate (Cat. No. 925804) was used for detection followed by hematoxylin counterstaining. Images were captured on a Keyence microscope with a 40X objective. Scale bar: 50 µm

    U-87 MG cells (panel A, negative cell line) and HaCaT cells (panel B, positive cell line) were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 100% ice-cold methanol for 10 minutes and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-Keratin 14 (clone W24068C), followed by incubation with Alexa Fluor® 594 Goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective and merged. Scale bar: 50 µm

    Whole cell extracts (15 µg total protein per lane) from the indicated cell lines resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-Keratin 14 (clone W24068C) overnight at 4°C. Description

Keratin 14 is a member of the keratin family, the most diverse group of intermediate filaments. This gene product, a type I keratin, is usually found as a heterotetramer with two keratin 5 molecules, a type II keratin. Together they form the cytoskeleton of epithelial cells. Mutations in the genes for these keratins are associated with epidermolysis bullosa simplex.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).

For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with any of the following:
-Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100
-Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol
-100% ice-cold methanol only

For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with any of the following:
-Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803)
-True-Phos™ Perm Buffer (Cat. No. 425401)
-True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) 
-True-Nuclear™ Transcription Factor Buffer Set gives the most robust signal.

This product does not cross-react with mouse keratin 14 by Western Blot.
