IHC staining with Purified anti-MRP-14 (S100A9) (clone A22135C) on formalin-fixed paraffin-embedded human esophagus tissue. Following antigen retrieval using 1X Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated without (panel A) or with (panel B) Purified anti-MRP-14 (S100A9) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured using with a 40X objective and merged. Scale bar: 50 µm -
Lysed human whole blood was fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes, permeabilized with 100% ice-cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-MRP-14 (S100A9) (clone A22135C), followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective and merged. Scale bar: 50 µm -
Lysed human whole blood gated on granulocyte population (positive expression, filled histogram) and lymphocyte population (negative expression, open histogram) were fixed and permeabilized using Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) and intracellularly stained with Purified anti-MRP-14 (S100A9) (clone A22135C), or Purified Mouse IgG2b, κ Isotype Control (Cat. No. 400301) (dashed histogram, representative of both the positive and negative cell populations) followed by incubation with PE Goat anti-mouse IgG (Cat. No. 405307).

The S100A8/A9 complex has been shown to play a role in calcium-dependent activation and exhibits antibacterial, antifungal, immunomodulatory, and anti-proliferative effects. It also functions as a chemoattractant for neutrophils and serves as an endogenous activator of Toll-like receptor 4 (TLR4). S100A9 has been identified as a marker of CD14⁺ HLA-DR⁻/low myeloid-derived suppressor cells (MDSCs).

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol.
For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803). Fixation/permeabilization with either True-Phos™ Perm Buffer (Cat. No. 425401) or True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) is not recommended.
