Whole cell extracts (15 µg total protein per lane) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-PIAS4 (clone W22136A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-β-actin (Cat. Description

PIAS4 (Protein Inhibitor of Activated STAT protein 4), also known as E3 SUMO-protein ligase PIAS4, is a multifunctional protein that regulates transcription and is essential in various cellular processes. PIAS4 facilitates the attachment of SUMO (Small Ubiquitin-like Modifier) to target proteins and modulates protein stability, localization, and interactions. PIAS4 regulates the activity of transcription factors such as STATs (Signal Transducers and Activators of Transcription) by promoting their SUMOylation. PIAS4 participates in the DNA damage response, where it facilitates the recruitment of repair factors like 53BP1 to sites of DNA damage. This SUMOylation-driven process is vital for preserving genomic stability, preventing mutations, and reducing the risk of tumorigenesis. PIAS4 also influences chromatin dynamics by interacting with chromatin-associated proteins, affecting chromatin structure and gene expression patterns. PIAS4 is also crucial in immune signaling, particularly in pathways like interferon signaling and NF-κB activation. By SUMOylating transcription factors and signaling molecules, PIAS4 fine-tunes the immune response, balancing pro-inflammatory and anti-inflammatory signals. This antibody is not recommended for immunocytochemistry (ICC) or intracellular flow cytometry (ICFC).
