Whole cell extracts (15 µg total protein per lane) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with Purified anti-TMEM97 (clone W22180E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin (Cat. No. 664804) was used as a loading control at a 1:5000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining with Purified anti-TMEM97 (clone W22180E) was performed on FFPE human pancreas tissue following antigen retrieval using 1X Citrate buffer (pH 6.0) (Cat. No. 420902). The tissue sections were incubated with Purified anti-TMEM97 (clone W22180E) (panel B and merged in panel C) followed by Alexa Fluor® 594 Goat anti-rat (Cat. No. 405422) (red) or secondary antibody only (negative control, panel A). Nuclei were counterstained with DAPI (Cat. No. 422801) (blue) (panels A and C). The image was captured wit a 40X objective. Sigma intracellular receptor 2 (σ2R), also known as SIGMAR2 or TMEM97, is a transmembrane protein that is associated with the endoplasmic reticulum (ER), membranes and lysosomes. It is widely distributed in normal tissues such as the brain, lungs, heart, skeletal muscles, testis, and ovary. TMEM97 has been demonstrated to regulate the cholesterol transporter NPC1 in maintaining cholesterol homeostasis. Elevated expression of TMEM97 is a potential prognostic biomarker for breast, lung, colon, ovarian, bladder and oral squamous cancers. TMEM97 is also implicated in several neurological disorders and age-related diseases such as Alzheimer's disease.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunohistochemistry on FFPE tissues (IHC-P), antigen retrieval with Citrate Buffer (Cat. No. 420901) is recommended.
This antibody (clone W22180E) does not cross-react with mouse TMEM97 in Western Blot.
We do not recommend this antibody for use in immunocytochemistry (ICC).
