Stability Testing for Recombinant Mouse VLDLR. Recombinant Mouse VLDLR was aliquoted in PBS, pH 7.2 at 0.2 mg/mL and one aliquot was kept at 4°C (Control), and another was frozen and thawed four times (4x Freeze/Thaw). After this procedure, the samples were tested for their ability in a binding assay with Recombinant Mouse LRPAP. The ED50 for this effect is 10 – 60 ng/mL.

Very Low-Density Lipoprotein Receptor (VLDLR) is a transmembrane protein involved in lipid metabolism and neuronal migration. Encoded by the VLDLR gene on chromosome 9 in humans, the sequence of VLDR comprises of approximately 875 amino acids. The protein contains a ligand-binding domain, an epidermal growth factor (EGF)-like domain, an O-linked sugar domain, a transmembrane region, and a cytoplasmic tail containing NPXY motifs essential for endocytosis. VLDLR is characterized by modular arrangement of its extracellular domains which enables binding to apolipoprotein E (ApoE)-containing lipoproteins and interactions with signaling molecules demonstrating its critical role in ligand recognition and endocytosis. VLDLR mediates the uptake of triglyceride-rich lipoproteins and is crucial in reelin signaling during neuronal migration in the developing brain. The receptor regulates lipid homeostasis and contributes to the organization of cortical layers. Mutations or dysfunctions in VLDLR are linked to VLDLR-associated cerebellar hypoplasia (VLDLRCH), a rare autosomal recessive disorder characterized by ataxia, intellectual disability, and cerebellar malformations. Additionally, VLDLR has been implicated in atherosclerosis, Alzheimer's disease, and other neurodegenerative conditions due to its involvement in lipid metabolism and signaling pathways.

- The 780 amino acid recombinant protein has a predicted molecular mass of approximately 86.4 kD. The DTT-reduced and non-reduced proteins migrate 75 - 130 kD by SDS-PAGE. The predicted N-terminal amino acid is Gly.

- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin the vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 μg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
