IHC staining with Purified anti-CD73 (clone W23165G) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval with 1X Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated without (panel A) or with (panel B) Purified anti-CD73 (clone W23165G) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged. Scale bar: 50 µm -
Whole cell extracts (15 µg total protein per lane) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-CD73 (clone W23165G) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

CD73 (cluster of differentiation 73) is a membrane-bound glycoprotein and ectoenzyme. To activate adenosine signaling CD73 catalyzes the hydrolysis of AMP generated by CD39 to adenosine. This role makes CD73 crucial to the regulation of purinergic signaling and immune homeostasis and response. CD73 also serves a variety of physiological functions including lymphocyte binding, epithelial ion transport, ischemic preconditioning, tissue-barrier function, T cell activation, and tissue homeostasis. Overexpressed in several cancers and implicated in tumor progression, metastasis, and the impairment of anti-tumor immune response via adenosine signaling, CD73 is a target of interest in cancer therapy. It is also a potential immunotherapy target for the treatment of pain, inflammation, and hypoxia.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 0.1 - 1.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 1.0 - 10.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
In Western Blot, this antibody does not cross react with mouse CD73.
This product is not recommended for use in immunocytochemistry (ICC) or intracellular flow cytometry (ICFC) due to dim signal.
