COLO205 (human colorectal tumor cell line) was stained with purified anti-human CA19-9 (clone 121SLE) (filled histogram) or purified mouse IgM, κ isotype control (open histogram) followed by anti-mouse IgM PE. -
Following antigen retrieval using 1X Tris-EDTA Buffer (Cat. No. 422704), formalin-fixed, deparaffinized human pancreas tissue sections were incubated with 5 µg/mL of purified anti-human CD99 (Cat. No. 324502) and purified mouse IgM, κ isotype control (Cat. No. 401602) (panel A) or purified anti-human CA19-9 (clone 121SLE) (panel B) overnight at 4°C. The tissue was then stained with rat anti-mouse IgM Alexa Fluor® 647 (red) (Cat. No. 406526) and rat anti-mouse IgG2a, κ Alexa Fluor® 594 (green) (Cat. No. 407120). Nuclei were counterstained with DAPI (blue) (Cat. No. 422801). Images were captured using a 10X objective. Scale bar: 50 µm

Carbohydrate Antigen 19-9 (CA19-9), also known as sialyl-Lewis A, is a cell surface glycoprotein complex. CA 19-9 expression requires the Lewis gene product, 1,4-fucosyltransferase, found only in patients with Le (a-b+) or Le (a+b-) blood groups. CA19-9 antigen is highly expressed in a variety of gastrointestinal epithelial malignancies including colon cancer and pancreatic cancer, and in breast cancer and small cell lung cancer, but has a limited expression on normal tissues.

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 1.0 µg per million cells in 100 µL volume. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
