Human peripheral blood lymphocytes were treated with (left) or without (right) 12 µg/mL of anti-human IgG, IgM antibody (Cat. No 397302) for 5 minutes. Cells were then fixed with Fixation Buffer (Cat. No. 420801), permeabilized with True-Phos™ Perm Buffer (Cat. No. 425401) and stained intracellularly with anti-human CD20 (clone 1412) FITC and anti-human BLNK Phospho (Tyr96) (clone A19008A) APC.

Description

B cell linker protein (BLNK), a member of the SLP-76 family, is an adapter molecule that plays important roles in B cell antigen receptor (BCR) engagement and B cell activation. Cross-linking of the BCR stimulates various downstream signaling pathways involved in cell proliferation and apoptosis. Intracellular protein tyrosine kinases such as Syk and Lyn have been implicated in BCR signal transduction and are thought to play an important role in B cell development. Phosphorylation of human BLNK at tyrosines 84, 178, and 189 (Y84, Y178, and Y189) creates docking sites for effector proteins GRB2, Vav, Nck and PLCγ2, which in turn leads to the activation of downstream signaling pathways. Phosphorylation of tyrosine 96 (Tyr96) facilitates BTK binding and plays a critical role in B cell maturation.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 µL per million cells in 100 µL staining volume or 5 µL per 100 µL of whole blood. It is recommended that the reagent be titrated for optimal performance for each application.
