Whole cell extracts (15 µg total protein per lane) from HAP1 cells with or without LDHA CRISPR knockout were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-LDHA (clone A22115F) overnight at 4°C.     IHC staining with Purified anti-LDHA (clone A22115F) on formalin-fixed paraffin-embedded human liver tissue. Following antigen retrieval using 1X Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated without (panel A) and with (panel B) Purified anti-LDHA (clone A22115F) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged.

    HAP1 cells (panel A) and HAP1 LDHA knockout cells (Horizon Cat. No. HZGHC004917c008, panel B) were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 100% ice-cold methanol for 10 minutes and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-LDHA (clone A22115F), followed by incubation with Alexa Fluor® 594 Goat anti-mouse IgG (red, Cat. No. 405326) for 1 hour at room temperature. Nuclei were counterstained with DAPI (blue, Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective and merged. Scale bar: 50 µm

Description

L-lactate dehydrogenase A chain, also known as LDHA, is an enzyme that plays a key role in cellular energy metabolism, particularly under anaerobic conditions. LDHA catalyzes the conversion of pyruvate to lactate, which coupled with the oxidation of NADH to NAD+ is essential for maintaining glycolysis when oxygen is limited. LDHA is widely expressed, with the highest levels in tissues with high energy demands such as muscle. In cancer cells, LDHA supports the Warburg effect by promoting lactate production, thus LDHA is a potential target for cancer therapy due to its role in tumor progression and metabolic reprogramming.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).

For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with any of the following:
- Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol
- 100% ice-cold methanol only
Note: Fixation/ permeabilization with Fixation buffer (Cat. No. 420801) followed by 0.5 % Triton X-100 is not recommended due to dim signal.

For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) or True-Phos™ Perm Buffer (Cat. No. 425401). Signal is brighter using True-Phos™ Perm Buffer. 
Note: Fixation/permeabilization with True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) is not recommended due to dim signal.
