Recombinant mouse IL-1RL1 (ST2)-Fc (Cat. No. 764902) (black circles) binds to immobilized recombinant mouse IL-33 (Cat. No. 580506). Ultra-LEAF™ purified anti-mouse IL-33 (clone A23126A) (purple squares) inhibits the binding in a dose-dependent manner, whereas the Purified Rat IgG2b, λ Isotype Control (Cat. No. 403801) (gray triangles) does not have the effect. This antibody blocks the binding of 0.5 µg/mL mouse IL-1RL1 (ST2)-Fc to 3 µg/mL immobilized recombinant mouse IL-33. ND50 range is 0.2 - 1.5 µg/mL. -
Recombinant mouse IL-33 (Cat. No. 580506) (black circles) induces proliferation of D10.G4.1 cells. Ultra- LEAF™ purified anti-mouse IL-33 antibody (clone A23126A) (purple squares) neutralizes the D10.G4.1 cell proliferation induced by recombinant mouse IL-33 at 0.2 ng/mL in a dose-dependent manner, whereas the LEAF™ purified rat IgG2b, λ isotype Control (gray triangles) does not have this effect. ND50 range is 0.8 - 6 µg/ml.

IL-33 belongs to the IL-1 family with close structural similarities with IL-18 and IL-1β. IL-33, IL-1β, and IL-18 are synthesized as biologically inactive precursor. These in-active precursors are released in their active mature form after undergoing cleavage by caspase-1 enzyme. Like IL-1β, IL-33 stimulates target cells by binding to the IL-1R/TLR superfamily member ST2 to activate NF-κB and MAPK pathways. In addition, IL-33 can also act as a nuclear factor, known as NF-HEV. Exhibiting transcriptional regulatory properties, NF-HEV been demonstrated to being abundantly expressed in the nucleus of high endothelial venules of lymphoid organs that are associated with chromatin.

Each lot of this antibody is quality control tested by blocking the binding of 0.5 µg/mL of mouse IL-1RL1 (ST2)-Fc (Cat. No. 764902) to immobilized 3 µg/mL recombinant mouse IL-33 (Cat. No. 580506). ND50 range is 0.2 - 1.5 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

High levels of mouse IL-33 mRNA are detected in the stomach, lung, spinal cord, brain, and skin. Expression within these organs is restricted to a few cell types, such as epithelial cells, fibroblasts, and smooth muscle cells. In addition, mouse IL-33 mRNA is found in resting dendritic cells and activated macrophages. Endothelial cells constitute a major source of IL-33 mRNA in chronically inflamed tissues.
- Function
- IL-33 drives production of Th2-associated cytokines from in vitro polarized Th2 cells. In mice, IL-33 injection induced the expression of IL-4, IL-5, and IL-13 and led to severe pathological changes in the lung and the digestive tract. In addition, IL-33 acts as a chemoattractant for Th2 cells, both in vitro and in vivo. TNF-α and IL-1β are activators of IL-33 transcription in fibroblasts and keratinocytes.
