Whole cell extracts (15 µg protein) from serum-starved cells treated without (-) or with (+) PMA (200 nM) for 4 hours were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane and probed with Purified anti-c-Fos (clone W23257D) overnight at 4°C. Proteins were visualized by chemiluminescence detection HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Lane M: molecular weight marker -
Serum-starved HeLa cells treated without (panels A and C) or with PMA for 4 hours (200 nM, panels B and D) were fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 100% ice-cold methanol for 10 minutes and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-c-Fos (clone W23257D), followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Cells were also stained with Alexa Fluor® 488 anti-Tubulin-α (Cat. No. 627906) (green) and nuclei were counterstained with DAPI (Cat. No. 422801) (blue). The images were captured and merged on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm -

The c-Fos protein, or FBJ murine viral (v-Fos) oncogene homolog, is a member of the Fos family of transcription factors. c-Fos forms heterodimers with proteins of the Jun family of transcription factors to form the AP-1 (Activator Protein-1) complex, which regulates critical cellular processes, including cell proliferation, differentiation, and survival. C-Fos has been demonstrated to play a critical role in angiogenesis, bone development, and in the progression of cancer. C-Fos is also often used as an indirect marker of neuronal activity.

ICC, ICFC - Verified - Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with either of the following:
- Fixation Buffer (Cat. No. 420801) with 0.5 % Triton X-100

For use in intracellular flow cytometry (ICFC), it is recommended to fixation/permeabilization with True-Phos™ Perm Buffer (Cat. No. 425401).
