Jurkat cells pulsed with 5-ethynyl-2′-deoxyuridine (EdU) for 3 hours (right) or untreated control cells (left) were stained using the Swift-Click™ Green EdU Kit and DAPI. -
Representative cell cycle analysis data of bone marrow cells harvested from a C57BL/6 mouse injected intraperitoneally with 1 mg EdU. The bone marrow was harvested 2 hours post-injection and stained with the Swift-Click™ Green EdU Kit and DAPI. -
Representative ICC staining using the Swift-Click Green EdU Kit. A549 cells were grown on coverslips in the presence of EdU (10 uM) for 2 hours and fixed with Fixation Buffer (Cat. No. 420801) for 30 minutes. Fixed cells were blocked with 3% BSA for 10 minutes, permeabilized using 0.5% Triton X-100 for 20 minutes, and then stained with Azide-Green cocktail (green) for 30 minutes at ambient temperature in the dark. Cells were then stained with 2.5 ug/mL of anti-PCNA Alexa Fluor® 594 (red) for 1 hour at ambient temperature in the dark. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801). The image was captured with a 40X objective.
The Swift-Click™ Green EdU Kit allows the rapid fluorescent detection of incorporated nucleoside analog in proliferating cells for cell cycle analysis. The staining procedure consists of labeling cells through incubation with nucleoside analog 5-ethynyl-2´-deoxyuridine (EdU), followed by cell fixation and permeabilization using our True-Nuclear™ Transcription Factor Buffer Set, and fluorescent EdU labeling. Fluorescent EdU labeling is catalyzed by Click chemistry, a Nobel Prize-recognized chemical reaction that rapidly links fluorophore-azide molecules to the alkyne group present in EdU in the presence of Cu2+ ions. This method bypasses radioactivity use, as well as long and harsh permeabilization and chromatin denaturation steps, allowing for a faster, simpler-to-follow procedure for cell cycle analysis.
