Human peripheral blood leukocytes were surface stained with anti-human CD4 (clone RPA-T4) APC and then fixed, permeabilized, and intracellularly stained with the True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401). Cells were then stained with anti-human FOXP3 (clone 206D) Brilliant Violet 421™ and anti-human EOS (clone W16032B) Alexa Fluor® 488 (left) or rat IgG2a, κ Alexa Fluor® 488 isotype control (right). Data shown was gated on the CD4+ lymphocyte population.

EOS also known as IKZF4 is an IKAROS Family Member expressed in lymphocytes, specifically in Tregs. It plays an important role in the inhibitory function of Tregs. It may be involved in the development of the central and peripheral nervous systems as well as lymphoid development.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular flow cytometry using our True-Nuclear™ Transcription Factor Staining Protocol. For flow cytometric staining, the suggested use of this reagent is 5 µL per 106 cells in 100 µL volume. It is highly recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 488 has a maximum emission of 519 nm when it is excited at 488 nm.
