Whole cell lysates (15 µg total protein per lane) from HeLa S3 cells untreated (-) or treated with Nocodazole to arrest the cell cycle at G2/M stage (+) were resolved by SDS-PAGE using 4-12% Bis-Tris gels. Then transferred to a PVDF membrane and probed with Purified anti-Rb Phospho (Ser795) (clone A24001E). Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607903) was used as a loading control. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. IHC staining with Purified anti-Rb Phospho (Ser795) on formalin-fixed paraffin-embedded human ovarian cancer tissue. After antigen retrieval with 1X Citrate Buffer (Cat. No. 420902), the tissue was either untreated (left panels) or treated with Lambda Phosphatase Protein (LPP) (right panels). The tissue was then incubated with Purified anti-Rb Phospho (Ser795) (clone A24001E) and subsequently with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322). Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured using a 40X objective, showing the Alexa Fluor® 647 channel alone (bottom images) or merged with the DAPI channel (top images). Scale bar: 50 µm -

The Retinoblastoma protein (Rb) is a tumor suppressor that regulates cell cycle progression from G1 phase into the S phase. This regulation is crucial for preventing uncontrolled cell proliferation, which can lead to cancer. Hence, mutations or dysfunctions in the RB1 gene are associated with several cancers including retinoblastoma, lung, bladder, and breast cancers. Phosphorylation at serine 795 of the Rb protein plays a crucial role in regulating its function. The serine 795 phosphorylation leads to Rb dissociation from E2F transcription factor and allowing E2F to induce the transcription of cell cycle genes. This regulatory mechanism is vital for normal cell function and its dysregulation can contribute to cancer development.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401). Similar results were also obtained using True-Nuclear™ Transcription Factor Buffer set (Cat. No. 424401) or Cyto-Fast™ Fix-Perm Buffer set (Cat. No. 426803).
For use of this antibody in Immunocytochemistry (ICC), we recommend using PBS-Tween-20 0.05% as a wash buffer. For use of this antibody in immunohistochemistry on formalin-fixed paraffinembedded tissues (IHC-P), we recommend antigen retrieval using Citrate Buffer (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704). - RRID
