What are ELISpot assays?

The Enzyme-Linked Immunospot (ELISpot) assay is a powerful immunological technique designed to detect and quantify cytokine-secreting or analyte-releasing cells at the single-cell level. It is based on the principles of the sandwich ELISA method, but extends its capabilities to identify individual analyte secreting cells among large populations. ELISpot is an invaluable tool for studying antigen-specific immune responses in areas such as vaccine development, infectious diseases, and cancer immunotherapy.

How ELISpot Works

 

The assay leverages plate-bound antibodies to capture analytes secreted by cells directly in their immediate environment. These captured analytes form complexes that are visualized using an enzymatic reaction, resulting in a localized, blue colored spot. Each visible spot corresponds to a single cell that secreted the analyte, hence the name "ELISpot."
